Immunophenotyping by flow cytometry has proven to be a valuable adjunct in the diagnosis and prognosis of various leukemias. Analysis of surface marker CD antigens by flow cytometry can distinguish benign from malignant leukocytosis and determine the cell lineage and stage of maturation. A clinical pathologist interprets each case.
The specimen requirement is one 5 mL green top (sodium heparin) tube of blood, one green top tube containing at least one mL of bone marrow, 10 mL of body fluid, or 1 g of tissue wrapped in saline dampened gauze and placed in a Petri dish with wet toweling. All specimens should be stored and transported at room temperature.
